Substrate is the nutrient-bearing material on which mushrooms are grown. It can be grain (such as rye or wild bird seed), coconut coir, vermiculite, straw, or manure-based mixes, depending on the cultivation method and species. Mycelium grows through the substrate during colonization, consuming its nutrients before mushrooms form and fruit.

How Substrate Choice Affects a Grow

Different substrates suit different stages and goals of cultivation. Sterilized grain is commonly used to prepare "grain spawn" — mycelium-colonized grain that acts as a vigorous starting culture — because grain provides a nutrient-dense, relatively uniform base that colonizes quickly under sterile conditions. Bulk substrates, often a mix of coir, vermiculite, and sometimes gypsum or manure, are used for the larger-volume fruiting stage; they are typically pasteurized rather than fully sterilized, since bulk substrate preparation is harder to keep sterile at scale and pasteurization is generally sufficient once it has been inoculated with already-colonized grain spawn.

Substrate composition and moisture directly affect yield, contamination risk, and how many flushes a substrate can support. A substrate at proper field capacity — holding as much water as it can without becoming waterlogged — supports healthy mycelial growth, while over-wet substrate is prone to bacterial contamination and under-hydrated substrate colonizes poorly and produces smaller fruits. Substrate type can also influence which species grow best: wood-loving species like Psilocybe cyanescens generally do better on wood chip or hardwood-based substrates, reflecting their natural role as lignicolous fungi, while many cultivated Psilocybe cubensis strains do well on standard grain-to-bulk setups.

Substrate selection is one of the first practical decisions in any cultivation method, and mismatches between substrate and technique (or species) are a common source of poor yields or failed grows, alongside contamination and inconsistent environmental control.
